to identify five different unknown mineral samples. [NOTE: cause & effect] the unknown lab report for my pure culture 18 i am in need of a great hypothesis for this subject for this lab 2 cultures e coli using ine slide for eahc sample gram staining procedure a slide is prepared by using sterileapply a primary stain (crystal violet), fix it with Gram's iodine (which fixes the primary dye inside the cell), decolorize with 95% ethyl alcohol to wash out the crystal violet-iodine (CVI) complex, and apply a counterstain, Safarin. Under a microscope, if the gram stain is purple, the bacterium is gram positive, if the stain is red, it is gram negative. The second procedure was the catalase test, this test is useful in distinguishing gram postive cocci from one another. On a slide a loopful of the unknown bacteria and 3 drops of hydrogen peroxide is mixed together to see if the bacteria produce the enzyme catalase. If the bacteria is catalase postive it will produce bubbles due to the release of oxygen , if the bacteria is negative for catalase there will be no bubbling. After these two procedures I was able to narrow down my unknown bacteria #18 to two bacteriums ( S.aureus and S.epidermis) because the gram stain was positive with purple grape-like clusters and the catalase test produce bubbles so therefore the bacteria was positive for catalase.
